Research

Atherosclerosis is the leading cause of cardiovascular disease and may lead to myocardial infarction, stroke or gangrene.   Risk  factors for atherosclerosis include hypertension, diabetes, hypercholesterolemia, cigarette smoking and may include infectious agents such as Chlamydia and cytomegalovirus (CMV). The inflammatory response to CMV and atherogenesis share many elements in common (see figure below).   Our research involves studying role of cytomegalovirus infection in atherogenesis and the complex relationship between CMV and pro-inflammatory cytokines within the blood vessel wall.  Monocyte chemoattractant protein-1 (MCP-1) is a chemokine produced by vascular endothelium that acts to direct monocytes to blood vessel walls following endothelial injury.

We have used MCP-1 transgenic mice to study the relationship between CMV infection and MCP-1.  Murine CMV infection accelerates inflammation in mouse tissues overexpressing MCP-1, while markedly reducing serum MCP-1 levels in MCP-1 transgenic mice.   MCMV has a large genome and has multiple molecular means of modifying the host inflammatory response. 

We are currently developing a double transgenic mouse (TIE2-rTA/TRE2-MCP-1) that will express MCP-1 in vascular endothelium and be doxycycline inducible.  This mouse model should help us determine how CMV interacts with MCP-1 and other pro-inflammatory cytokines to initiate or exacerbate vascular disease and eventually lead to new ways to treat or prevent atherosclerosis.
