ResearchThe overall goal of our research program is to study the interaction of growth factors and ovarian hormones on the natural history of mammary cancer development. Much of experiments utilize genetically engineered mice which express transforming growth factor alpha (TGFalpha) in the mammary gland. These mice have a high incidence of mammary tumor development. In addition the tumors which develop in TGFalpha mice represent a special type of clinical breast cancer: those which express estrogen receptor but which do not any longer express progesterone receptor. This clinical breast cancer type is often refractory to antiestrogen therapy so our TGFa mouse model may prove useful in dissecting the mechanism of antiestrogen resistance. In addition, we use breast cancer cells in culture to determine the role of signal transduction cascades in promoting cell survival and proliferation. Techniques used include: histological evaluations of tissues and gene expression analysis using microarray technology (Affymetrix) and quantitative PCR. In addition, total and phosphoprotein levels are measured using immunohistochemistry and western blotting. Finally, in vitro cell cycle dynamics are evaluated by flow cytometry.
