ResearchThe focus of our research is on the biochemical and molecular mechanisms by which foreign chemicals including pharmaceuticals, pesticides, environmental pollutants, and industrial compounds interact with biological systems to cause adverse, toxic tissue damage. We are particularly interested in the mitochondrion as a primary intracellular target of chemical-induced toxicity. In addition to studying bioenergetic poisons, we are also investigating the mechanistic basis of metabolic diseases caused by exposure to chemicals that interfere with the molecular biology of the mitochondrion. This includes interference with mitochondrial DNA replication and biogenesis, expression of the mitochondrial genome, mitochondrial protein synthesis, and assembly and incorporation of the individual respiratory complexes in the inner mitochondrial membranes. Implicit to these investigations is identifying and understanding the regulation of transcription factors responsible for the coordinated expression of the nuclear and mitochondrial genomes. We believe that mitochondrial biogenesis is a well-coordinated, multifaceted and dynamic process that is essential to the adaptation of the cell to changing metabolic environments and that interference with this compensatory mechanism underlies the bioenergetic disorders associated with assorted chemically-related toxicities.
