Dr. Miller completed his Pediatric residency (2000) and Pediatric Endocrinology fellowship (2003) at the Mayo Clinic. He has been an Assistant Professor in Pediatric Endocrinology since 2003. He selected Douglas Yee, MD, an Established Investigator in the CHRCDA, as his mentor. Dr. Miller’s research centers on the biology of glioblastoma multiforme, a very aggressive tumor with high mortality rates. Previous studies have shown a role for the IGF system in the growth and survival of multiple malignant cell types, including brain tumors. Recently developed reagents and strategies for inhibition of IGF-1 signaling, such as selective tyrosine kinase inhibitors, monoclonal antibodies and small inhibiting mRNAs, provide novel methods for analyzing IGF action and represent potential treatments to inhibit tumor growth. Dr. Miller uses U87 glioblastoma cells as a model of brain tumor growth in vitro and in vivo. Based upon this proposed research, Dr. Miller received the Lawson Wilkins Pediatric Endocrine Society Novo Nordisk Clinical Scholar Award for 2004-2006. Dr. Miller has demonstrated that several inhibitors of IGF signaling inhibit U87 cell growth in vitro. Current studies involve examination of the ability of IGF signaling inhibitors to impair tumor growth in vivo using U87 tumor xenografts in immune-deficient mice. Future studies will analyze the efficacy of combining inhibitors of IGF signaling with radiation and chemotherapy agents in vitro and in vivo.
